CD70 Molecular Characteristics & Key Mutations

CD70 (also known as CD27 ligand) is a member of the tumor necrosis factor superfamily (TNFSF7). It contains a TNF homology domain (THD; UniProt P32970) and forms a native trimeric structure critical for receptor binding. Two key mutations in the LPFS3 region have been documented: variant VAR_081988 leads to loss of protein expression, while VAR_081989 preserves expression but abolishes interaction with CD27.
